Biggest risk

The IPF failure removes a pipeline expansion opportunity and increases reliance on Revuforj execution and cGVHD readouts. BofA cut its target from $26 to $23 while maintaining Buy; Stifel reportedly maintained Buy and a $40 target, indicating divergent post-result interpretations. https://www.tipranks.com/news/the-fly/syndax-price-target-lowered-to-23-from-26-at-bofa-thefly-news https://uk.investing.com/news/stock-market-news/stifel-maintains-syndax-stock-rating-after-trial-failure-93CH-4908166

What would change my mind?

Revuforj growth and the remaining cGVHD program could support recovery despite the lost IPF opportunity. Q2 Syndax revenue was $72.8 million, including $54.7 million of Revuforj revenue and $18.1 million of Niktimvo collaboration revenue—not the full $60.3 million of Niktimvo product sales reported by partner Incyte. Niktimvo product sales are recorded by Incyte; Syndax recognizes collaboration revenue based on its share of net commercial profit, so product sales cannot be added directly to Syndax revenue.

Commentary

The packet's $13.95 October 13 close was about 15.7% below the prior close following the October 12 MAXPIRe announcement; Dow Jones also reported an immediate after-hours decline. BofA lowered its target, while another reported rating and target were unchanged. The numerical news-tone estimate is qualitative; zero-valued social, options, short-interest and employee fields mean unavailable, not neutral. Low coverage and incomplete analyst-revision data limit conviction. https://www.marketscreener.com/news/syndax-says-pulmonary-fibrosis-drug-trial-misses-primary-endpoint-ce785dddd880f127 https://www.tipranks.com/news/the-fly/syndax-price-target-lowered-to-23-from-26-at-bofa-thefly-news

Frontline cGVHD Phase 2 readout
Syndax expects Phase 2 data for axatilimab plus ruxolitinib in frontline chronic graft-versus-host disease in Q4 2026. This is a distinct clinical test, not a reversal of the IPF result. [#SEC-8K-2026-08-04] https://www.sec.gov/Archives/edgar/data/1395937/000119312526332736/sndx-ex99_1.htm

The Phase 2 MAXPIRe trial missed its primary forced-vital-capacity endpoint, and Syndax will stop developing axatilimab for IPF. Further repricing remains possible, although the company says its other axatilimab trials are unaffected. Q2 Revuforj revenue rose 91% year over year to $54.7 million, with prescriptions up 15% sequentially. Sustained treatment duration and new-patient starts are the next tests of commercial momentum. Q2 R&D expense rose to $68.0 million from $62.2 million a year earlier, primarily from frontline AML trials; continued investment and potential dilution matter while Syndax remains loss-making. https://syndaxpharmaceuticalsinc.gcs-web.com/news-releases/news-release-details/syndax-reports-second-quarter-2026-financial-results-and
